报告内容:In plant, primary transcripts (pri-miRNAs) are processed into ~21nt long miRNAs by trimeric DCL1-SE-HYL1 processing machinery. Here, we present the crystal structures of Arabidopsis SERRATE and HYPONASTIC LEAVES1 (HYL1). In vitro pull-down assay showed that SE provides the platform for HYL1 and DCL1 binding, whereas in vitro RNA binding and in vivo mutant rescue experiments suggested that the conserved non-canonic C2H2 zinc finger domain coupled with C-terminal unstructured tail recognizes the structural features of miRNA precursors to assist the precise processing by DCL1. Structural and functional study demonstrated that HYL1 binds to the miRNA/miRNA* region of miRNA precursors as a dimer to work together with SE and DCL1 for miRNA processing.
